Intended use For In Vitro diagnostic use.
This product is intended for use as a diluent in the preparation of primary antibodies and negative control
reagents. It is to be used with antibodies and/or antigens that yield high background in immunohistochemical
(IHC) staining procedures.

Reagent provided 0.05 mol/L Tris-HCl buffer containing 0.1% Tween, stabilizing proteins to reduce background, and 0.015 mol/L
sodium azide.
Precautions 1. For professional users.
2. This product contains sodium azide (NaN3), a chemical highly toxic in pure form. At product concentrations,
though not classified as hazardous, build-ups of NaN3 may react with lead and copper plumbing to form
highly explosive metal azides. Upon disposal, flush with large volumes of water to prevent azide build-up in
plumbing.1,2
3. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin.
4. Unused solution should be disposed of according to local, State and Federal regulations.
Reactivity For demonstration of tissue antigens by IHC, optimal staining performance is dependent on the correct dilution
of the primary antibody. The interaction between antibodies and epitopes consists mainly of relatively weak
forces such as van der Waals forces, electrostatic forces and hydrophobic forces, which are influenced by pH
and ionic strength. Consequently, to assure optimal performance of the antibody with the least amount of
background, the choice of diluent is important.
Applications This product provides an appropriay buffered solution for the dilution of both polyclonal and monoclonal
antibodies and for the preparation of negative control reagents in IHC. This product is particularly useful for
reducing background staining while maintaining specific reactivity. Antibody Diluent with Background Reducing
Components can also be used to stabilize diluted antibodies when stored at 2–8°C. This diluent should be
brought to room temperature and mixed thoroughly prior to use.
Limitations Tissue staining results are dependent upon the handling and processing of the tissue prior to staining.
Aldehyde-containing fixatives, such as formalin and glutaraldehyde frequently contribute to increased
background staining. Improper fixation, freezing, thawing, washing, drying, heating or sectioning may produce
artifacts, antibody trapping or false-negative results. Also, false-positive staining may be caused by crossreactivity
of other IHC staining reagents to e.g. endogenous alkaline phosphatase, endogenous peroxidase,
pseudoperoxidase, endogenous avidin-binding activity and/or by nonspecific reaction with necrotic or
degenerated cells.
Storage Store at 2–8°C. Do not freeze.
